It is told that the project team of the “Stem Cell-based Kidney Structure and Function Restoration”, aimed at solving the practical problem of donor kidney shortage in clinical practices, have made several breakthroughs in a series of core technological innovations by building a stem cell-based regenerative medicine system to restore the structure and function of the kidney: firstly, based on various stem cells, hiPSC-derived organoids, and the proprietary extracorporeal circulation perfusion culture system, they have established a screening and identification system for kidney cells with differentiation potential. Secondly, by adopting acellular kidney scaffolds of rats, pigs, and humans, they have built the implantation system of mature and stable kidney cells with differentiation potential. Thirdly, they have built a rat kidney orthotopic transplantation model with acellular scaffolds, and completed a systematic evaluation of the function of the restored kidney. Through the above systematic innovations, four key technologies have been developed, an integrated scientific and technological innovation chain of basic research - product development - clinical application has been refined, and related technologies have been promoted in five cooperating organizations. Six projects have been approved by the National Natural Science Foundation of China, one laboratory has been titled the provincial key laboratory for kidney restoration, 28 excellent SCI papers on relevant topics have been published, recording a total impact factor of 76.748, and two postdoctoral fellows, 12 doctors and 18 masters have been cultivated. The project has created outstanding social benefits, and has made important contributions to supporting and serving the continuous improvement of national and local scientific and technological innovation capabilities, promoting economic and social development continuously, and catering to the goals of “Healthy China and “Healthy Shandong”.
